Relatively new music box programmer and extremely novice musician here. In my experience, there is no clear step-by-step process. Use what tools you have to make what you are trying to do work. To go straight from an audio file (mp3, etc.) to a melody is nearly impossible (to my knowledge). I have either been finding midi files to either import directly in or manually edit in a daw before importing. Another method I have begun doing is to find sheet music and copy it over into a music notation program (I'm using MuseScore Studio) then export the midi file from there.

The tools mentioned by Tuxedo_Mask are great. In my experience, I've found that ai-midi is more reliable and accurate than the other two tools though. If you specifically want to convert piano audio to midi (which I'm guessing you do if you want a melodie box song), then eldoraudio's piano audio to midi tool is excellent: eldoraudio.com/products/piano-audio-to-midi
